摘要: 目的建立活体可视化胰岛β细胞发育的立体表达动物模型。方法利用分子克隆、显微注射、全胚胎原位杂交和活体摄像等方法,建立绿色荧光蛋白标记胰岛β细胞的转基因斑马鱼。结果建立并筛选获得胰岛β细胞发育的转基因斑马鱼,证实荧光蛋白与内源性胰岛素的共定位表达,实现活体动态监测胰岛β细胞的发育情况。结论胰岛β细胞的转基因斑马鱼动物模型实现在整体动物水平上研究胰岛β细胞的发育。

Abstract: Objective we have established pancreatic β-cell developmet fish model, which showed specific spatial expression patterns in vivo. Methods molecular clone, microinjection, whole embryo in-situ hybridization(WISH)and fluorescence microscopy in living were used to analyze of β-cell development by generation of transgenic zebrafish. Results screening and established pancreatic β cells of transgenic zebrafish, then confirmed that fluorescence protein expression in the same spatiotemporal pattern with endogenous insulin gene to achieve dynamic monitoring islet β-cell development situation in vivo. Conclusion the pancreatic β cells of transgenic zebrafish animal model can be successfully traced pancreatic β cell development in whole level.
