蜡块表面脱钙法在骨肉瘤标本制片中的应用

Application of wax block surface decalcification in the preparation of osteosarcoma specimen

摘要: 目的 探讨蜡块表面脱钙法(SD)对骨肉瘤组织形态学及分子生物学方面的影响,建立新的骨组织脱钙方法。方法 收集17例骨肉瘤标本,每例切取含肿瘤骨组织2块,分为表面脱钙组(SD)和常规脱钙组(RD),石蜡切片分别行HE染色;免疫组织化学检测 ki-67、stab2、caspase9、survivin、和BAX表达;MDM2探针荧光原位杂交染色检测MDM2基因的扩增。结果 表面脱钙法与常规脱钙法HE染色2者无差异;抗原和MDM2基因检测SD组高于RD组(P<0.05)。结论 蜡块表面脱钙法在组织形态学、抗原及核酸保存方面优于常规脱钙法。

Abstract: Objective To investigate the effects of wax surface deca-lcification on osteosarcoma morphology and molecular biology. Met-hods 17 specimens of osteosarcoma were collected, Two pieces of bone tissue containing tumor were cut out and divided into two gr-oups: surface decalcification group and routine decalcification group,the paraffin sections were stained with HE; Immunnohistochemical Staining was used to detected the expression of Ki-67,statb2,caspase9, su-rvivin and BAX; Fluorescence in situ hybridization with MDM2 probe was used to detected the gene amplification; Results There was no difference between the surface decalcification group and the routine decalcification group on HE staining,The differenceof antigen expression and MDM2 gene amplification was significant Conclusions The surface decalcification of wax block is superior to the routine decalcification in morphology,antigen and preservation of antigen and nucleic acid.
