FGF9和CHIR99021诱导胚胎干细胞形成肾脏样结构

FGF9 and CHIR99021 induce embryonic stem cells forming kidney-like structure

摘要: 目的 在体外诱导胚胎干细胞(ESCs)向肾脏细胞分化,并自发形成包含肾小球、近端小管、远端小管以及血管网络的肾脏样结构。方法 在体外将ESCs培养至40%~50%汇合度。然后在成纤维细胞生长因子9(FGF9)与肝糖原合成激酶3β受体的选择性抑制剂CHIR99021的作用下,诱导ESCs分化形成肾脏样结构,对该肾脏样结构分别进行免疫荧光染色检查与电镜检查,以观察其内部结构并且成像记录。结果 通过细胞因子FGF9与CHIR99021的作用,成功诱导ESCs分化形成了肾脏样结构,结构鉴定证实其中有肾小球、近端小管、远端小管以及血管网络的存在。结论 利用ESCs分化可以形成包含完整肾单位及血管网络的肾脏样结构,其结构类似于正常肾脏。

Abstract: Objective To induce embryonic stem cells (ESCs) to differentiate into the renal cells in vitro and to spontaneously formed the kidney-like structure including glomerular, proximal tubules, distal tubules and vascular network. Methods ESCs were cultured to 40-50% of confluence. Under the action of cytokine FGF9 and CHIR99021, the differentiation of ESCs was induced to form the kidney-like structure, which was examined by immunofluorescence staining and electron microscopy to observe its internal structures and take images. Results Through the action of cytokine FGF9 and CHIR99021, ESCs can be induced to differentiate into renal like structures, including glomerular, proximal tubules, distal tubules and vascular network, which was confirmed by the structural identification. Conclusions The kidney-like structure generated by the differentiation of ESCs contains complete renal units and vascular network, which is similar to the normal kidney.
