急进高原后外周血单个核细胞内相关易感基因表达与急性高原反应分析

The analysis between AMS and the correlated predisposing genes of ascentting of plateau in mononuclear cells of peripheral blood

摘要: 目的 探讨低氧诱导因子1(HIF-1α)、P38MAPK和血管内皮生长因子(VEGF)在急进高原后外周血单个核细胞的表达变化,以及其与急性高原反应(AMS)的关系。方法 对某部队赴青海省玉树抗震的官兵分别于进入高原前后48 h采集外周血,提取单个核细胞用RT-PCR法检测P38MARK及HIF-1α mRNA表达,Western blot法检测HIF-1α及VEGF蛋白表达。按照《急性高原反应的诊断和处理原则》诊断AMS。结果 与进入高原前相比,HIF-1α mRNA和蛋白表达水平及p38MARK mRNA表达水平均明显增高(p<0.001); VEGF蛋白水平也上调(p<0.01);HIF-1α及VEGF蛋白表达水平与AMS的发病程度高度相关,相关系数分别为0.875和0.675(p<0.001)。结论 HIF-1α、P38MARK和VEGF在急进高原48 h后表达水平均显著增加,HIF-1α和VEGF蛋白表达水平与AMS发病呈显著正相关。

Abstract: Objective To explore the change of expression of hypoxia inducing factor 1(HIF-1? ), P38MARK and vascular endothelial growth factor (VEGF )after ascenting of plateau in vivo, and the correlation with the pathogenesy of AMS. Methods Collecting soldiers' venous blood before they asent of plateau and after they arrive in Yushu in Qinghai province 48 h later, extracting mononuclearcell the expression of mRNA of HIF-1? and P38MARK were detected by RT-PCR and the expression of protein of HIF-1? and VEGF was detected by Western blotting. The AMS is diagnosed according to《The Diagnose And Processing Principle Of AMS 》 Results After asentting of plateau the expression levels of mRNA of HIF-1? , P38MARK and the protein of HIF-1? and VEGF all increase significancely(p<0.01). At the same time, the change of expression levels of HIF-1? and VEGF protein are highly corellated with AMS , the coefficient correlation is separately 0.806 and 0.675(p<0.001). Conclusion The expression level of HIF-1?, P38MARK and VEGF increase significantly after asentting of plateau, especially the changes of HIF-1? protein and VEGF, it is highly correlated with the extent of AMS.
