TLK2促进人乳腺癌细胞系增殖、迁移和侵袭

TLK2 promotes the proliferation, migration and invasion of human breast cancer cell lines

摘要: 目的 探究Tousled样激酶2 (TLK2)对乳腺癌细胞的影响及其可能的机制。方法 用免疫组织化学和Western blot检测TLK2在乳腺癌组织和细胞系中的表达。用RT-qPCR检测乳腺癌组织和细胞系中TLK2 mRNA以及miR-622的表达。用CCK-8、BrdU和Transwell小室法检测TLK2过表达质粒和miR-622抑制剂对HCC70细胞增殖、迁移和侵袭的影响。通过Targetscan在线分析数据库和双荧光素酶报告基因分别预测和验证miR-622和TLK2之间的靶向调控关系。结果 乳腺癌组织或细胞系中的TLK2表达水平显著上调 (P<0.05)。TLK2显著促进HCC70细胞增殖、迁移和侵袭 (P<0.01)。可能的作用机制表明miR-622可结合TLK2的3′ UTR。结论 TLK2参与促进HCC70细胞增殖、迁移和侵袭,且被miR-622负向调控。

Abstract: Objective To explore the effects of TLK2 in breast cancer (BC) cells and its possible mechanism. Methods Immunohistochemistry and Western blot were used to detect the expression of TLK2 in BC tissues and cell lines. RT-qPCR was employed to detect the expression of TLK2 mRNA and miR-622 in BC tissues and cell lines. CCK-8, BrdU and Transwell assays were used to detect the effects of TLK2 over-expression plasmid and miR-622 inhibitors on the proliferation, migration and invasion of HCC70 cells. The targeted regulatory relationship between miR-622 and TLK2 was predicted and verified by Target scanning online analysis database and dual-luciferase reporter gene, respectively. Results The expression of TLK2 in BC tissues or cell lines was significantly up-regulated (P<0.05), and TLK2 significantly promoted the proliferation, migration and invasion of HCC70 cells (P<0.01). The possible mechanism suggested that miR-622 can bind to the 3′UTR of TLK2. Conclusions TLK2 is involved in promoting the proliferation, migration and invasion of BC cells, and is negatively regulated by miR-622.
