淫羊藿苷通过下调转移相关蛋白2抑制MGC803细胞的增殖、迁移和侵袭

Icariin inhibits proliferation, migration and invasion of MGC803 cell line by down-regulating metastasis-associated protein 2

摘要: 目的 探讨淫羊藿苷通过调控转移相关蛋白2(MTA2)表达抑制人胃癌细胞系MGC803细胞增殖、迁移和侵袭的作用机制。方法 以不同浓度淫羊藿苷干预MGC803细胞,MTT法检测细胞活力;Transwell和Western blot分别检测细胞迁移和侵袭及MMP-2、MMP-9和MTA2蛋白表达;将MTA2 siRNA转染至MGC803细胞后检测检测细胞活力、迁移和侵袭;将pcDNA-MTA2 转染至MGC803细胞联合淫羊藿苷处理,MTT和Traswell实验检测细胞活力、迁移和侵袭。结果 不同浓度淫羊藿苷均能有效抑制MGC803细胞活力。以200μmol/L淫羊藿苷干预MGC803细胞后,细胞迁移和侵袭能力下降(P <0.05),MMP-2、MMP-9、MTA2蛋白表达下调。敲减MTA2能够抑制细胞活力、迁移和侵袭。过表达MTA2可部分逆转淫羊藿苷对MGC803细胞活力、迁移和侵袭的抑制作用。结论 淫羊藿苷能够通过调控MTA2、MMP-2和MMP-9蛋白表达,抑制胃癌细胞增殖、迁移和侵袭。

Abstract: Objective To investigate the mechanism of icariin inhibiting proliferation, migration and invasion of MGC803 cells by regulating MTA2. Methods Cell viability of MGC803 cells after treatment with multiple concentrations of icariin was detected by MTT assay. Migration, invasion and the protein expression of MMP-2, MMP-9 and MTA2 in MGC803 cells were detected by Transwell and Western blot, respectively. Transfection MTA2 siRNA into MGC803 cells, and detecting cell viability, migration and invasion. Cell viability, migration and invasion of MGC803 cells after transfection of pcDNA-MTA2 combined with icariin were detected by MTT assay and Traswell, respectively. Results Icariin at different concentrations could effectively inhibit viability of MGC803 cells. Migration, invasion and the protein expression levels of MMP-2, MMP-9 and MTA2 of MGC803 cells after treatment with 200μmol/L icariin were decreased. Knockdown MTA2 can inhibit cell viability, migration and invasion. Overexpression of MTA2 partially reverses the inhibitory effect of icariin on the viability, migration and invasion of MGC803 cells. Conclusions Icariin can inhibit proliferation, migration and invasion of gastric cancer by regulating the expression of MTA2, MMP-2 and MMP-9.
