HPV16E7在宫颈癌组织中表达升高并促进宫颈癌细胞系HeLa和C33A的增殖

HPV16E7 is up-regulated in cervical cancer tissues and promotes the proliferation of cervical cancer cell lines HeLa and C33A

摘要: 目的 研究乳头瘤病毒HPV16E7在宫颈癌中表达水平及其在宫颈癌细胞HeLa和C33A中的作用机制。方法 实时荧光定量PCR(RT-qPCR)和免疫组织化学染色检测宫颈癌患者组织及其对照组中HPV16E7、CEA和CA125 mRNA和蛋白水平,并分析3者之间的关系。RT-qPCR 和ELISA实验检测宫颈癌患者血清及其对照组中HPV16E7、CEA和CA125水平变化。Western blot、MTT、集落形成、流式细胞计量术周期和凋亡实验探究HPV16E7在HPV阴性细胞系C33A及HPV阳性细胞系HeLa中发挥的作用。结果HPV16E7、CEA和CA125 mRNA及蛋白水平在宫颈癌患者组织和血清中是高表达的(P<0.05)。HPV16E7与CEA和CA125具有正相关的关系(P<0.05)。HPV16E7在C33A细胞中不表达,在HeLa细胞中高表达。HPV16E7能够促进宫颈癌细胞系C33A和HeLa细胞的增殖能力,促进细胞周期进程,并能够抑制细胞凋亡。结论 HPV16E7能够促进宫颈癌细胞的恶性行为,其表达水平在宫颈癌患者血清和组织中显著升高,并与CEA和CA125呈正相关的关系。

Abstract: Objective To investigate the expression level of HPV16E7 in cervical cancer and its’ mechanism in HeLa and C33A cell lines. Methods RT-qPCR and Immuohistochemical staining assays were used to detect the expression level of HPV16E7, CEA and CA125 in cervical cancer tissues and analyzed the correlation of HPV16E7 and CEA, CA125. RT-qPCR and ELISA assay were used to detect the expression level of HPV16E7, CEA and CA125 in cervical cancer serum. Western blot, MTT, colony formation, cell cycle and cell apoptosis assay were used to investigate the role of HPV16E7 in C33A (HPV-) and HeLa (HPV+) cells. Results The mRNA and protein levels of HPV16E7, CEA and CA125 in cervical cancer tissues and serum were up-regulated(P<0.05). The correlation of HPV16E7 and CEA, CA125 was positively relevant(P<0.05). HPV16E7 was up-regulated in HeLa cells and no expression in C33A cells. HPV16E7 promoted proliferation, cell cycle progress and inhibited cell apoptosis in HeLa and C33A cells. Conclusions Up-regulated HPV16E7 promotes malignant behavior in cervical cancer, which is positively related to CEA and CA125.
