人端粒酶反转录酶过表达抑制低氧/复氧大鼠心肌细胞的凋亡

Overexpression of human telomerase reverse transcriptase inhibits myocardial apoptosis induced by anoxia/reoxigenation in rats

摘要: 目的 探讨人端粒酶反转录酶(TERT)在大鼠心肌低氧/复氧(A/Ro)损伤中的保护作用。方法 利用腺病毒将端粒酶催化亚基转染至大鼠心肌细胞;利用慢病毒介导小干扰核糖核酸(siRNA)沉默心肌细胞TERT基因;研究TERT过表达和沉默后对大鼠心肌细胞A/Ro诱导的细胞凋亡的影响。结果 A/Ro组细胞凋亡率显著高于对照组;TERT过表达组可显著降低A/Ro心肌细胞的凋亡率;过表达的TERT可降低心肌细胞CytC的表达并增强P21的表达(P<0.05)。结论 过表达的TERT可抑制A/Ro诱导的心肌细胞凋亡,其机制可能与调控p21及CytC基因表达有关。

Abstract: Objective To investigate the Effects of human telomerase reverse transcriptase on myocardial apoptosis induced by Anoxia/Reoxigenation in rats. Methods Adenovirus mediated transfection of TERT gene into myocardial cells of neonatal rats, lentivirus-mediated siRNA silencing the TERT. To investigate the protective effects of human telomerase reverse transcriptase on myocardial Anoxia/Reoxigenation injury in rats. Results The apoptosis rate of A/Ro group was significantly higher than that of the control group. Compared with the A/Ro group, TERT over-expression group could significantly reduce the rate of apoptosis, and over-expression of TERT decreased the expression of CytC gene and increased the expression of P21 gene in cardiomyocytes(P<0.05). Conclusions Over expression of telomerase reverse transcriptase inhibits myocardial apoptosis induced by A/Ro. The mechanism is related to the regulation to the expression of P21 and CytC genes.
