筋脉通含药血清对高糖培养大鼠雪旺细胞β-catenin、GSK-3β及髓鞘零蛋白表达的影响

Effects of Jinmaitong serum on the expression of β-catenin, GSK-3β and myelin protein zero in Schwann cells of rats cultured in high glucose medium

摘要: 目的 探讨不同浓度筋脉通含药血清对高糖培养大鼠雪旺细胞(SCs)β-catenin、GSK-3β及髓鞘零蛋白(P0)的影响。 方法 SD大鼠灌胃获得含药血清及正常血清,原代培养雪旺细胞,随机分为正常对照组、高糖组、筋脉通5、10和20倍干预组,72 h后分别用CCK、real-time PCR和Western blot法检测各组SCs细胞增殖活性及β-catenin、GSK-3β和P0的mRNA与蛋白表达。结果 与正常组相比,高糖组SCs细胞增值率显著降低(P<0.01),β-catenin和P0 mRNA与蛋白表达显著降低(P<0.01),而GSK-3βmRNA表达显著上升(P<0.05)。中药筋脉通干预后使高糖培养的SCs细胞β-catenin、P0的mRNA及蛋白表达显著回升(P<0.01),而GSK-3βmRNA表达显著回降(P<0.01),且与浓度相关。结论 高糖可引起SCs细胞损伤,中药筋脉通可以通过调节β-catenin、GSK-3β水平抑制该作用。

Abstract: Objective To investigate the effect of serum containing different concentrations of Traditional Chinese medicine Jinmaitong onβ-catenin、GSK-3βand P0 in Schwann cells cultured in high glucose medium. Methods Twenty-five male Sprague-Dawley rats were randomly treated with 5, 10, 15 and 20 times of Jinmaitong and distilled water. Schwann cells were divided into six groups, which are Control group, High glucose group, 5 times group, 10 times group, 15 times group, 20 times group. 72 hours later, the proliferative activity of SCs cells were detected by CCK, the mRNA and protein expression of β-catenin、P0 and GSK-3β was detectived by rt-PCR and Western blotting. Results High glucose medium could inhibit the proliferation of Schwann cells, down-regulate the expression of β-catenin and P0(P<0.01), and up-regulate the expression of GSK-3β(P<0.05) significantly. But Jinmaitong can invert the results (P<0.01, P<0.05). Conclusion High glucose medium will injure the proliferation of Schwann cells, But Jinmaitong can increase the proliferation activity of Schwann cells, and promotes the secretion of P0 Partially dependent on up-regulating the activity of β-catenin and down-regulating the activity of GSK-3β.
