甲醛对鼠BM-MSCs细胞系的遗传毒性影响

Genotoxic effect of formaldehyde on mouse BM-MSCs

摘要: 目的 研究甲醛对BM-MSCs的遗传毒性,从而为甲醛导致白血病的发生提供生物学依据。方法 采用噻唑蓝比色(MTT)法检测BM-MSCs的增殖活性;用彗星、KCl-SDS沉淀、姐妹染色单体互换(SCE)和微核(MN)检测甲醛对BM-MSCs的遗传毒性效应。结果 75~200 μmol/L甲醛可呈剂量依赖抑制BM-MSCs的增殖(P<0.01),诱导BM-MSCs DNA断裂效应呈现先增高后降低趋势,在125 μmol/L时,达到峰值,其中75~150 μmol/L甲醛作用较对照组显著升高(P<0.01);甲醛在≥125 μmol/L时可以明显引起BM-MSCs DNA-蛋白交联(DPCs)、SCE和MN的形成,较对照组显著升高(P<0.01)。结论 甲醛可抑制BM-MSCs的增殖活性,并且对BM-MSCs具有一定的遗传毒性效应。

Abstract: Objective Study the genotoxic effect of formaldehyde on BM-MSCs, thus will provides the biological evidence for the formaldehyde to lead to leukemia. Methods Proliferation activity was measured by MTT assay; Genotoxic effect was detected by comet assay, KCl-SDS precipitation assay, sister chromatid exchange (SCE) test and micronucleus (MN) test. Results 75~200 μmol/L formaldehyde can inhibit the BM-MSCs proliferation in a dose-dependent manner (P<0.01); DNA strand breakage increased gradually at increasing concentrations below 125 μmol/L, however, when formaldehyde concentration was over 125 μmol/L, DNA strand breakage decreased. There were significant rises in 75~150 μmol/L formaldehyde groups compared with control group (P<0.01); Induction of DNA-protein crosslinks (DPCs), SCE and MN in ≥125 μmol/L formaldehyde groups were significant higher than in control group (P<0.01). Conclusion Formaldehyde can inhibite the BM-MSCs proliferation activity, and have a genotoxic effect on the BM - MSCs.
