橙皮苷预处理减轻Con A致小鼠急性肝损伤

Hesperidin pretreatment attenuates concanavalin A-induced acute liver injury

摘要: 目的 研究橙皮苷(HDN)预处理对刀豆蛋白A(Con A)致小鼠急性肝损伤的保护作用及机制。方法:给小鼠连续灌胃HDN 10d,用Con A诱导小鼠急性肝损伤,计算肝和脾指数,测定小鼠血清丙转氨酶(ALT)和谷草转氨酶(AST)的水平,测定肝匀浆丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性,HE染色观察肝脏病理变化,TUNEL法检测肝细胞凋亡,RT-PCR法检测小鼠肝组织Bcl-2和Bax mRNA表达。结果HDN预处理能降低Con A致急性肝损伤小鼠肝和脾指数(P<0.01);降低血清中ALT和AST水平(P<0.01);降低MDA含量(P<0.01),提高SOD活性(P<0.01);明显抑制肝细胞凋亡;上调Bcl-2表达(P<0.01),下调Bax表达(P<0.01)。结论HDN预处理对Con A致小鼠急性肝损伤具有保护作用,可能与其抗脂质过氧化作用及抑制肝细胞凋亡有关。

Abstract: Objective To investigate the protective effects of hesperidin (HDN) on hepatic injury induced by concanavalin A (Con A) in mice. Methods Hesperidin was administrated to mice daily for ten days via gastric gavage and then acute liver injury model was established by intravenous administration of Con A. Liver and spleen were weighed to calculate liver and spleen index. ALT and AST in serum, MDA and SOD was measured by reverse transcription polymerase chain reaction (RT-PCR). Results HDN pretreatment significantly decreased liver and spleen index, serum levels of ALT and AST (P<0.01); pathological changes in hepatic tissue were dramatically improved; the content of MDA was decreased and the activity of SOD was increased (P<0.01); the extent of hepatic apoptosis was significantly reduced; more importanty, Bcl-2 mRNA expression was up-regulated while Bax mRNA expression was down-regulated markedly (P<0.01). Conclusion HDN pretreatment protects mice from Con A-induced acute liver injury probably through inhibiting lipid peroxidation and hepatocyte apoptosis.
