MEF来源的间充质干细胞诱导生成的调节型树突状细胞的基因表达谱分析

The gene expression profile analysis of regDC induced by MEF-MSC

摘要: 目的 研究MEF来源的间充质干细胞(MEF-MSC)诱导的调节型树突状细胞(MEF-regDC)与不成熟树突状细胞(imDC)和成熟树突状细胞(maDC)之间的基因表达谱差异,从而揭示前者的基因表达特点。方法 将骨髓造血干细胞与MEF-MSC共培养制备MEF-regDC,用Agilent芯片对3种树突状细胞(DC)进行基因芯片检测及相关分析,分析项目包括:差异倍数计算、主成分分析(PCA)和Go分类。 结果 MEF-regDC具有特征性的形态、表型和基因表达谱,PCA分析表明,MEF-regDC、imDC和maDC是不同的DC亚群,MEF-regDC与imDC相比,表达差异在2倍以上的基因数共有13 369个,差异在6倍以上的有3 247个,MEF-regDC与maDC相比,表达差异在2倍以上的基因数共有11 326个,差异在5倍以上的有3 299个,进一步的分析显示,3者在功能调节相关基因,免疫相关的基因和信号通路相关的基因的表达也有显著差异。 结论 MEF-MSCs诱导HPCs生成的MEF-regDC是一不同于传统imDC及maDC的具有特征性基因表达谱的DC亚群。

Abstract: Objective The aim is to investigate the difference of gene expression between MEF-regDC and imDC & maDC, in order to demonstrate the character of MEF-regDC. Methods We first harvest MEF-regDC by co-culture of bone marrow hematopoietic stem cell with MSC, then analysis 3 types of DCs, including Fold change, Principal Component Analysis and GoTerm to Gene. Results MEF-regDC has the characteristic of morphology, phenotypes and gene expression profile, the PCA illustrate that MEF-regDC and imDC & maDC are 3 different type of DC, and there are 13369 genes that are 2 folds different between MEF-regDC and imDC, and the number of genes that are 6 folds different between the 2 types of DCs is 3247; meanwhile, the number of different genes between MEF-regDC and maDC are 11326 and 3299, respectively. Further analysis demonstrate that, there are obvious difference of function regulate genes, immune-related genes and signaling pathways related genes among these three type of DCs. Conclusion MEF-regDC is a new type of DC which is different from imDC and maDC, and has characteristic gene expression profiles.
