金黄色葡萄球菌分泌趋化抑制蛋白与C5aR,C5L2的相互作用

The interaction between C5aR,C5L2 and chemotaxis inhibitory protein secreted by Staphylococcus aureus

摘要: 目的 探讨金黄色葡萄球菌分泌的趋化抑制蛋白(CHIPs)与G蛋白偶联受体(GPCR) C5a受体(C5aR)或促酰化蛋白受体(C5L2))的相互作用。 方法 将纯化的CHIPs蛋白分别与表达C5aR, C5L2, CXC 趋化因子受体(CXCR3)的人肾胚胎上皮细胞293T分别孵育,用Western blot检测结合情况。 结果 CHIPs蛋白与C5aR相结合,与C5L2和CXCR3无明显结合。 结论 C5aR是CHIPs蛋白的受体,提示C5L2与C5aR在和CHIPs的结合作用上存在明显差异。

Abstract: Objective To investigate the physical association between G protein-coupled receptors (GPCR) including C5a receptor (C5aR), C5a Like Receptor 2 (C5L2) and chemotaxis inhibitory protein (CHIPs) secreted by Staphylococcus aureus. Methods The purified CHIPs was incubated with HEK 293T cells overexpressing C5aR, C5L2 and C-X-C chemokine receptor type 3 (CXCR3). Binding signal was detected by Western-blotting. Results HEK 293T cells overexpressing C5aR can efficiently bind to CHIPs. No apparent association between CHIPs and C5L2 or CXCR3 was detected. Conclusion C5aR not C5L2 is a membrane receptor for binding CHIPs, It suggesting a difference between C5aR and C5L2 in association with binding CHIPs.
