Nitric oxide inhibits cell proliferation and expression of C-FOS and C-JUN in gastric cancer cell line SGC-7901

摘要: 目的 探讨一氧化氮(nitric oxide, NO)对胃癌细胞SGC-7901增殖的影响及其机制。方法 MTT法测定细胞的生长,RT-PCR和Western印迹法检测C-FOS 和C-JUN的mRNA和蛋白表达。结果 SNP抑制SGC-7901细胞的生长(P < 0.05),并呈剂量和时间性降低C-FOS 和C-JUN mRNA及蛋白表达。结论 NO可能通过下调C-FOS 和C-JUN的表达而抑制胃癌细胞的增殖。

Abstract: Objective This study was to investigate the effect of nitric oxide (NO) on cell proliferation in gastric cancer cell line SGC-7901, and to investigate the relevant mechanisms. Methods MTT assay was used to detect the inhibitory effect of NO on cell growth. RT-PCR was used to detect the mRNA expression of C-FOS and C-JUN. Western blot was applied to study the protein expression of C-FOS and C-JUN. Results SNP inhibits the proliferation of SGC-7901 cells (P<0.05). SNP dose- and time-dependently decreased the expression of C-FOS, C-JUN mRNA and protein. Conclusions NO inhibits the proliferation of SGC-7901 cells, which is probably through NO down-regulated C-FOS and C-JUN expression.
