海马内注射鸢尾素缓解小鼠抑郁样行为

Intrahippocampal injection of irisin ameliorates depression-like behaviors in mice

摘要: 目的 探讨鸢尾素对抑郁症模型小鼠抑郁样行为的影响。方法 将 C57BL/6 小鼠随机分成模型组和健康组,模型组采用开放空间强迫游泳 (OSFS) 方法建立抑郁小鼠模型,造模成功后将造模组随机分成“模型-对照”组和“模型-鸢尾素”组。健康野生小鼠正常饲养,同样随机分为“健康-对照”组和“健康-鸢尾素”组。所有鸢尾素组小鼠通过脑立体定位注射向两侧海马分别注射77 pmol鸢尾素,对照组注射同体积的溶剂 (PBS)。利用强迫游泳及悬尾检测等实验对其抑郁样行为进行检测。结果 与健康组比较,抑郁模型组小鼠在强迫游泳测试 (P<0.01) 及悬尾测试 (P<0.05) 中的不动时间显著延长,证明成功模拟了抑郁症患者的绝望样行为,模型构建成功。与“模型-对照”组相比,鸢尾素可显著缩短“模型-鸢尾素”组小鼠在强迫游泳实验 (P<0.001) 及悬尾实验 (P<0.05) 中的不动时间。结论 鸢尾素可显著改善开放空间强迫游泳抑郁模型小鼠的抑郁样行为。

Abstract: Objective To investigate the effects of irisin on the depression-like behaviors in depression mice. Methods C57BL/6 mice were randomly divided into model and healthy groups. Mice in the model group were trained by open-space forced swim (OSFS) model and then were sub-divided into “model-control” and “model-irisin”. Healthy wild mice were bred normally and sub-divided into “healthy-control” and “healthy-irisin”groups. All mice in test group and in control groups were injected with 77 pmol irisin or the solvent (PBS) into both hippocampi by brain stereotactic injection, respectively. Depression-like behaviors were examined by forced swim test (FST) and tail suspension test (TST). Results Compared with the healthy ones, the model mice showed a markedly prolonged immobility time in FST(P<0.01) and TST(P<0.05), which indicated that the successfully established model can simulate the despair-like behaviors of patients with depression. Compared with the “model-control”, irisin can remarkably reduce the immobility time of “model-irisin” mice in FST(P<0.001) and TST(P<0.05). Conclusions Irisin can significantly improve the depression-like behaviors of OSFS depression mice.
