高血压视网膜病变患者外周血单个核细胞Notch1水平及其与血管内皮功能的相关性

Notch1 level and its correlation with vascular endothelial function of peripheral blood monocytes in patients with hypertensive retinopathy

摘要: 目的 探讨高血压视网膜病变(HRP)患者外周血单个核细胞Notch1表达情况及其与血管内皮功能的关系。方法 选择HRP患者42例为研究对象,另以同期健康体检人群30例作为对照组,收集外周血血清、单个核细胞,通过RT-PCR测定Notch1 mRNA水平,免疫印迹测定Notch1蛋白水平,测定血清VEGF、IGF-1、ET-1、NO水平,并进行统计学分析。结果 HRP组Notch1的mRNA(P<0.01)、蛋白水平(P<0.05)均明显低于对照组,Ⅲ期、Ⅳ期患者Notch1的mRNA、蛋白水平下降更为明显(P<0.01,P<0.05);HRP患者VEGF、IGF-1、ET-1明显高于对照组(P<0.05),NO明显低于对照组(P<0.05);Notch1与VEGF、IGF-1、ET-1呈显著负相关(P<0.05),与NO呈显著正相关(P<0.05);Notch1低水平是HRP发病的独立危险因素(OR:0.733,95% CI:0.601~0.835,P<0.001)。结论 HRP患者外周血单个核细胞Notch1水平下降,血管内皮功能紊乱可能与低Notch1相关。

Abstract: Objective To investigate the expression of Notch-1 in peripheral blood monocytes in patients with hypertensive retinopathy (HRP) and the possible relationship with vascular endothelial function. Methods Forty-two HRP patients were selected as HRP group and 30 healthy peoples were selected as control group. Peripheral blood serum and monocytes were collected. The levels of Notch1 mRNA and protein were determined by RT-PCR and Western blot, respectively. The level of VEGF, IGF-1, ET-1 and NO in serum was determined. Results The level of Notch1 mRNA and protein in HRP group was significantly lower than those in control group(P<0.01,P<0.05), especially in stage Ⅲ and Ⅳ(P<0.01,P<0.05). The levels of VEGF, IGF-1 and ET-1 in HRP group were significantly higher than those in control(P<0.05) and NO was significantly lower than that in control(P<0.05). The level of Notch1 was negatively correlated with VEGF, IGF-1 and ET-1(P<0.05) and positively correlated with NO in patients with hypertensive retinopathy(P<0.05). Low level of Notch1 was an independent risk factor for HRP (OR:0.733,95% CI:0.601-0.835,P<0.001). Conclusions The level of Notch1 of peripheral blood monocytes is decreased in HRP patients, and vascular endothelial dysfunction may be associated with low level of Notch1.
