Abstract: Objective To investigate the effects of seizure and/or an antiepileptic drug Carbamazepine (CBZ) on expression of P-glycoprotein (P-gp) encoded by a multidrug resistance gene. Methods Female SD rats were intracerebroventricularly injected with kainate (KA) and/or orally administered with CBZ or PBS. The animals were deeply anesthetized and perfused 3 d, 5 d, 7 d, 14 d, 21 d and 28 d after seizure or 7 d, 14 d, 21 d and 28 d after CBZ administration, and their brain was removed. Immunohistochemistry and double lebeling immunostaining were used for semiqualification analysis and location of P-gp in the brain, and Nissl staining was used to observe neuronal alterations in the hippocampus. Results The changes in EEG and behavior in the model rats were the same as those in human epilepsy. CBZ was weakly against the seizure generated by KA. Five days after seizure, P-gp expression in the hippocampus began to increase significantly and reached to its peak level at 7 d (P<0.001), then returned to the control level 21 d later. The immunopositive products were mainly present in the capillary endothelia in the hippocampus and also in the endothelia and neurons in the cortex. CBZ interference showed no effects on expression of P-gp compared with those in the PBS-injected rats. The changes in P-gp expression in KA+CBZ group were the same as those observed in the KA model rat, however, its P-gp expression at 7 d was stronger than that in the KA group. Conclusions Seizure may induce over-expression of P-gp, and CBZ may be involved in up-regulation of P-gp expression in the KA model rat.

Abstract:Objective To investigate the inhibitive effect of antisense VEGF165 cDNA on growth and angiogensis of human neuroblastoma in nude mouse models. Methods Three kinds of SH-SY5Y cells which had been stably transfected by sense VEGF165 cDNA, antisense VEGF165 cDNA and empty vector respectively, were inoculated subcutaneously to nude mice. The size of subcutaneous tumors was measured, and the morphology of tumor tissue was observed by microscope. The microvessel density (MVD) in tumor mass was analyzed by CD31 immunohistochemistry staining. Apoptotic cells were detected by the electron microscope. Results The growth of tumor mass, which was inoculated with antisense VEGF, decreased significantly in nude mice compared with empty vector group (P<0.05), while the growth rate oftumor in the sense VEGF group increased significantly in nude mice (P<0.05). The morphologic investigation of SH-SY5Y cells in antisense group showed blood vessel sparsity, smaller nuclei, fewer cytoplasm, lesser mitotic figure compared with the empty vector group. The electron micrographs of SH-SY5Y cells in antisense group showed the characteristics of apoptosis: the chromatin condensed in masses at the periphery of nuclei, and the nuclear fragmentation were seen, but they were not seen in sense group and empty vector group. MVD in rumor mass of the antisense group decreased significantly compared with empty vector (P<0.01), and MVD of sense group increased significantly (P<0.01). Conclusions Antisense VEGF165 cDNA transfection could inhibit the proliferation of SH-SY5Y cells in the nude mice, decrease angiogensis and increase tumor cells apoptosis.

ABSTRACT: Objective To prepare human rBPI23 and acquire its specific polyclonal antibody by immunizing rabbits, so that lay the foundation for preparation of its monoclonal antibody and later setting up immunological testing method for BPI. Methods PBV220-synBPI600 expressing vectors were transferred into competent cell, and the recombinant protein was expressed as inclusion body by temperature control. Its molecular weight was certified by SDS-PAGE, and antigenic specificity was analyzed by Western-blot. Antiserum against rBPI23 was acquired by immunizing rabbits, and the polyclonal antibody was purified by saturated ammonium sulphate precipitation. The titer of antibody was tested by indirect ELISA , and the specificity of antibody was analyzed by Western-blot. Results The recombinant protein was expressed mainly as inclusion body in Ecoli DH5α, SDS-PAGE showed that its molecular weight is 23 kD, which is consistent with the previous theoretical expection. The recombinant protein was analyzed by western-blot with rabbit polyclonal antibody against human BPI, and found that it could be recognized specifically by this polyclonal antibody. High titer of polyclonal antibody(1:320000) was acquired by immunizing rabbits, western-blot analysis revealed that these polyclonal antibodies could specifically react with rBPI23 and human standard sample of BPI. Conclusion Human rBPI23 was successfully prepared, and high titer of polyclonal antibody against human rBPI23 was acquired by immunizing rabbits.

Abstract:To construct pEGFP-Kallikrein eukaryotic expression vector which used green fluorescence protein as reportor gene, and transfect rabbit endothelial progenitor cells. The human tissue kallikrein(tHK)gene were amplified with PCR, and inserted into pEGFP-N3 vector. The rabbit endothelial progenitor cells(EPCs) were transfected with the formed plasmid by means of lipidosome, and the kallikrein-EGFP fused protein was visued directly with fluorensce microscope, and the expression of tHK was detected by RT-PCR(reverse transcription-PCR) and ELISA. Plasmids was formed correctly and detected by PCR meanwhile being sequenced, and expressed in the rabbit EPCs. The fused protein has actitivites of both kallikrein and EGFP. The tHK genetically modified EPCs could play a role in the treatment of artery injury and endothelial dysfunction.

AIM: This study was to ascertain the role of NF κB in cyclosporine A( CsA)-induced nephrotoxicity and relations between TGF β1,caspase3 and apoptosis . Methods: 64 wistar male rats were randomly divided into 4 groups of sixteen rats as follows. Vehicle,PDTC,CsA,CsA + PDTC. rats received a daily subcutaneous injection of CsA 15 mg.kg-1.d-1 ,and a daily subcutaneous injection of CsA 15 mg.kg-1.d-1 and PDTC 100mg.kg-1.d-1. BUN,serum creatinine, N-acetyl-beta-d-glucosaminidase (NAG) and urine routines were examined. The kidneys were processed for light microscopy,immunocytochemistry and TENUL. Results: A rat model of CsA-induced nephrotoxicity was established observed from light microscopy. CsA groups significantly increased the values of the BUN, SCr and urine NAG compared with the controls whereas CsA + PDTC group not. Immunohistochemistry revealed that the expressions of protein NF κB, TGF β1, caspase3 increased significantly in CsA-treated rats compared with conrtol ,whereas the expressions of protein NF κB, TGF β1, caspase3 was inhibited by PDTC. Apoptosis is observered in the CsA-induced chronic nephrotoxicity by TUNEL, PDTC blocked the apoptosis caused by CsA. Conclusions: CsA-induced chronic nephrotoxicity mainly include injuries of tubules,accompanied vacuolation, nucleus pycnosis and destroyed structures ,glomerulus injury also can be observed. Apoptosis is induced in the CsA-induced chronic nephrotoxicity and caspase3 may be involved in the regulation of this apoptosis,Inhibition of NF κB blockaded apoptosis in CsA-induced chronic nephrotoxicity . CsA induced up-regulated the expression of TGF β1 and significantly reduced the expression with PDTC. It is suggested that NF κB plays important roles in progression of chronic CsA nephrotoxicity and regulates other factors

Abstract Objective: To elucidate the features of the expression of p53, proliferation and apoptosis in gestational trophoblastic tumor (GTT). Methods: 10 cases of normal placenta, 10 cases of complete hydatidiform mole (CM), 20 cases of invasive hydatidiform mole (IM) and 19 cases of choriocarcinoma(CCA) paraffin-embedded tissues were studied. P53 and PCNA were detected with immunohistochemistry and apoptosis was detected with terminal deoxy-nucleotidyl transferase-mediated dUTP-biotin nick end labeling (TUNEL). Results: In NP, CM, IM and CCA, The p53 index(p53-I) in them were 4.12%, 21.68%, 39.61% and 27.39% respectively with significant differences among them (F=6.687, P=0.001). the PCNA index (PI) were 10.40%, 20.76%, 53.60% and 51.95% respectively with significant differences among them (F=17.510, P=0.000). The Apoptosis index (AI) in them were 1.88%, 2.59%, 6.45% and 1.26% respectively with significant differences among them (F=7.020, P=0.000). Significantly positive correlation was found between p53-I and PI(r=0.587,P=0.000), while there is no correlation between p53-I and AI(r=0.113,P=0.096). Conclusion: The process of GTT is associated with trophoblastic cell overproliferation because the proliferation and apoptosis are out of control. The increasing expression of muta-p53 contributes to the cell proliferation in GTT and can be helpful to the progress of GTT. No correlation was found between p53-I and AI.

Objective To investigate the effects of Rhodiola herb on renal tubulointerstitial injury in ADR rats.Methods The ADR model was established in rats by tail vein injection of adriamycin 3.0mg/kg twice .The rats were randomly as signed to normal control group,nephropathy group and Rhodiola herb treatment group.The rats were killed at the time of 4,8,12,16,20 week and to obtain samples.Renal histopathological changes were observed,the expressions ofα-SMA in the constit of the kidney Was examined by utilizing immunohistochemistry. Results Compared with the treatment group,the staining of α-SMA within the tubulonterstitium of nephropathy group was stronger.The expression of in interstitial α-SMA was increased with the degree of tubulointerstitial injury. Conclusion Rhodiola herb has a protective effect on tuhulointerstitial injury in ADR nephropathy,which may realize through the inhibition of epithelial-mesenchymal transition.
