Abstract: Objective To establish target SARS-CoV spike neutralizing antibody test model. Methods Construction of optimal whole SARS-CoV spike gene plasmid to induce mouse neutralizing antibody. Pretreatment with S318-510 protein targeting ACE2 to observe the inhibition change of SARS-CoV S pseudoviruses infection process via fluorescence microscope. Results SARS-CoV S1190 Abs can not only neutralize SARS-CoV pseudovirus entry into HEK293E/ACE2-Myc cells but also pretreatment with S318-510 bypass neutralization of SARS-S entry by double check. Conclusions This test model confirmation of SARS-CoV RBD targeting SARS-CoV spike neutralizing antibody may provide and explore test model of lethal viruses in laboratory without biosafety level 3.
