Liraglutide inhibit high glucose-induced TNF-a and ICAM-1 expression in rat glomerular mesangial cells

Abstract: Objective To explore the effect of Tumor necrosis factor-a(TNF-a) and intercellular adhesion molecule-1 (ICAM-1) in cultured rat glomerular mesangial cells ( RGMC) by high glucose and intervention of Liraglutide. Methods Cultured HBZY21 RGMCs were divided into 6 groups: control cells,high glucose stimulated cells,Liraglutide low、median and high( 10、100、1000nmol/L) concentration incubated cells stimulated with high glucose,PDTC intervention cells. The expressions of TNF-a and ICAM-1 of each group were measured by ELISA and RT-PCR. Results The level of TNF-a and ICAM-1 protein or mRNA was remarkably increased by high glucose,and Liraglutide could prevent this change . Compared with high glucose stimulated cells, the expression of TNF-a and ICAM-1 protein or mRNA was decreased with PDTC intervention cells(P<0.05).Conclusions Liraglutide might play an important role in preventing diabetes nephropathy by partly relieving the expression of TNF-a and ICAM-1 induced by high glucose.
