摘要

目的 研究曼陀罗属植物洋金花(Datura metel Linn.)干燥花的化学成分。方法 洋金花的干燥花经体积分数95%和70%乙醇加热回流提取,浓缩得到浸膏,用蒸馏水混悬,经石油醚、二氯甲烷和乙酸乙酯系统萃取,二氯甲烷和乙酸乙酯萃取部位经过硅胶柱、ODS柱、Sephadex LH-20凝胶柱和高效液相色谱制备等方法进行分离,再通过理化性质和NMR等技术方法对分离得到的化合物进行结构鉴定。结果 从洋金花中分离鉴定了13个化合物,分别是:dehydrovomifoliol(1),blumenol A(2),(6R,7E,9R)-9-hydroxy-4, 7-megastigmandien-3-one(3),oleracone B(4),黑麦草内酯(isololiolide, 5),山柰酚-3-O-β-D-葡萄糖苷(kaempferol 3-O-β-D-glucopyranoside,6),山柰酚-3-O-β-D-半乳糖苷(kaempferol 3-O-β-D-galactoside,7),山柰酚-7-O-α-L-鼠李糖苷(kaempferol 7-O-α-L-rhamnoside,8),七叶内酯(esculetin, 9),syringaresinol(10),2, 6-bis (1-phenylethyl) phenol(11),4-O-methylhonokiol(12)和(9E)-8,11, 12-trihydroxyoctadecenoic acid methyl ester(13)。结论 化合物1~5,10~13为首次从曼陀罗属中分离得到,化合物7和9为首次从洋金花中分离得到。

Abstract

OBJECTIVE To study the chemical constituents from the dry flowers of Datura metel Linn.. METHODS The dried flowers of D. metel Linn. were refluxed and extracted by 95% and 70% ethanol, concentrated to get the extract, suspended with distilled water, and then extracted by petroleum ether, dichloromethane and ethyl acetate. The dichloromethane and ethyl acetate extracts were isolated by silica gel column, ODS column, Sephadex LH-20 column and Pre-HPLC, respectively. Their structures of the isolates were elucidated on the basis of spectroscopic data including physical and chemical properties and NMR. RESULTS Thirteen compounds were respectively isolated and identified as follows: dehydrovomifoliol (1), blumenol A (2), (6R, 7E, 9R)-9-hydroxy-4, 7-megastigmandien-3-one (3), oleracone B (4), isololiolide (5), kaempferol 3-O-β-D-glucopyranoside (6), kaempferol 3-O-β-D-galactoside (7), kaempferol 7-O-α-L-rhamnoside (8), esculetin (9), syringaresinol (10), 2, 6-bis (1-phenylethyl) phenol (11), 4-O-methylhonokiol (12), and (9E)-8, 11, 12-trihydroxyoctadecenoic acid methyl ester (13). CONCLUSION Compounds 1-5, 10-13 are reported from the genus Datura for the first time, and compounds 7 and 9 are isolated from D. metel Linn. for the first time.
