Abstract��
OBJECTIVE To investigate the effects of sodium aescinate on the oxidize low-density-induced increases in nitric oxide (NO) production and the expression of inflammatory factors in the U937 cells.METHODS U937 cells were divided into 5 groups: normal group, ox-LDL group (U937 cells were incubated with 40 mg��L-1 ox-LDL for 24h), treatment groups with sodium aescinate (5, 10, 20 ��g��mL-1)( U937 cells were incubated with sodium aescinate and 40 mg��L-1 ox-LDL for 24 h). Then the production of NO in medium was measured by nitrate reductase. The expressions of tumor necrosis factor-�� (TNF-��) and interleukin-6(IL-6) in medium were detected by enzyme linked immunosorbent assay (ELISA). RESULTS In the ox-LDL group, the production of NO and the expression of TNF-�� and IL-6 were significantly increased compared with normal group. However, sodium aescinate (20,10 ��g��mL-1) treatment blocked ox-LDL-induced increases in NO, TNF-�� and IL-6. CONCLUSION Sodium aescinate is could attenuate the NO production, expressions of TNF-�� and IL-6 induced by ox-LDL in U937 monocyte cells, according to suppresses inflammatory reactions.
