Abstract��
OBJECTIVE To investigate hemolysis caused by the interaction between rabbit erythrocytes and cucurbitacin B galactosylated liver-targeting liposomes prepared with different adjuvants. METHODS The hemolytic phenomena of the three cucurbitacin B galactosylated liver-targeting liposomes formulations were observed by macroscopic observation and the optical microscope photos of the rabbit erythrocytes were taken with digital microscope. After being treated with a proper emulsion-breaker�� the hemolysis percentage of rabbit erythrocytes induced by liposomes were spectrophotometrically determined at 575 nm. RESULTS The three formulations of cucurbitacin B galactosylated liver-targeting liposomes with different adjuvants did not show any hemolytic behavior. The hemolysis percentage of the three formulations were all below 5% by using 10%Triton X-100 as the emulsion-breaker. CONCLUSION Three cucurbitacin B galactosylated liver-targeting liposomes formulations are injectable. The union of macroscopic observation�� the optical microscope observation and the spectrophotometry are reliable and feasible to evaluate the hemolytic behavior. 10%Triton X-100 can be used in the hemolysis test as a good emulsion-breaker for liposomes.
