摘要

目的 建立高效液相色谱法测定血浆中葫芦素B浓度的方法,研究葫芦素注射剂在小鼠体内的药动学。方法 采用高效液相色谱法,色谱柱:Hypersil ODS C18, 流动相:甲醇-水(72:28),筛选雌酚酮作内标。样品经萃取纯化后进样,检测波长228 nm,按内标法定量测定。结果 标准曲线在0.5~15μg·mL-1内有良好线性关系(r=0.9962),方法回收率在94.11%~99.77%,检测限4 ng,小鼠血浆中最低检测浓度为50 ng·mL-1。日内RSD小于12.12%,日间RSD小于15.63%。葫芦素水针剂小鼠静脉给药后体内的药-时过程符合二室模型。结论 首次建立的高效液相色谱法测定葫芦素B血药浓度的方法简便,快速准确,灵敏度高,适用于测定血浆中葫芦素B浓度及体内的药动学研究。

Abstract

OBJECTIVE The pharmacokinetics of cucurbitacins in mice was studied, and the HPLC method was established for the separation and quantitative determination of cucurbitacins B in plasma.METHOD Column: Hypersil ODS C18 (150 mm×4.6 mm, 5 μm); column temperature: 30℃;mobile phase: a mixture of methanol(MeOH) and water(72∶28); flow rate: 0.5 mL·min-1; detection wavelength: 228 nm; aliquot injected into the HPLC system: 20 μL.RESULTS The recovery of the assay was 94.11%~99.77% with the intra-day and inter-day RSD less than 12.12% and 15.63%,respectively.The detection limit was 4ng. The lowest detection concentration was 50 ng·mL-1 in plasma.CONCLUSON This method was found to be simple, rapid ,sensitive and accurate for the determination of CuCB in plasma.
