Abstract��
OBJECTTIVEThe correlation between the dissolution rate and the Aera Under Curve (AUC) of plasma concertration-time plot of gemfibrozil capsules was studied,the formulation of gemfibrozil capsules was selected in order to enhance its bioavailability. METHDOS��The dissolution of five batches of gemfibrozil capsules produced by two factories were determined by ultravilet spectrophotometery.The plasma concentrations of gemfibrozil capsules in 12 rabbits were determined by HPLC assay after an oral dose of 54.5 mg��kg-1.Their relative bioavailability were studied.The correlation between the mean cumulative dissolution rate in vitro- and AUC- in vivo- was analysed.The composition and processing of gemfibrozil capsules were selected with uniform design. RESULTS The difference of T d of two factories was significant (-P-��0.01)��One compartment model was fitted to the plasma concentration-time profiles.The relative bioavailiability was 71.69��(-P-��0.05).The correlation between their cumulative dissolution rate and AUC- was significant(-P-��0.01).The formulation of gemfibrozil capsules was composed of Gemfibrozil,Starch,Ls-HPC and CMS-Na. CONCLUSION The dissolution rate in vitro- of gemfibrozil capsules had a large influence on its bioavailability in vivo- and there was significant positive correlativity between them.The bioavailability of the gemfibrozil capsules was enhanced.
