Abstract��
OBJECTIVE: To explore the separation mechanism and the usage principle of silica gel-column quasi-ionexchange liquid chromatography for the analysis of alkaloids. METHODS:Orthogonal experiment was performed to do multifactors and multilevel test, and the retention time of different types of alkaloids was taken as an objective.Subsequently, the liquid chromatography analysis was made by using the mobile phase of different proportions of thanol-acetic acid-ammonium acetate. RESULTS: The concentration of ammonium acetate in the mobile phase had the greatest effect on retention time, and the next was acetic acid and thanol. CONCLUSION: The silica gel-column could be used not only in common positive phase chromatography, but also is the -Si-OH group at the surface of silica gel for ionexchange analysis.
