摘要

目的 建立高效液相色谱法同时测定八味檀香散中丁香酚和甘草酸含量的方法。 方法 采用Agilent ZORBAX Eclipse SB- C18 (4.6 mm×150 mm,5 μm色谱柱。流动相为乙腈-5%乙腈(含3%冰醋酸(27∶73;流速 1 mL·min -1,检测波长程序:0~17.0 min为280 nm,17.0~30 min 为250 nm。 结果 丁香酚和甘草酸保留时间分别约为15 和21 min,与各自相邻峰的分离度均大于1.5。以峰面积对进样质量浓度 (μg·mL-1 线性回归,丁香酚回归方程:Y=0.106 4ρ-1.338,r=0.999 9,线性范围77.76 ~777.6 μg·mL-1。甘草酸回归方程:Y=0.133 9ρ-0.222 0,r=0.999 9,线性范围14.40 ~144.0 μg·mL-1。丁香酚和甘草酸的回收率分别为99.0%和97.3%、RSD分别为1.5%和1.8%。 结论 本法操作简便,测定结果准确,重复性好,可用于八味檀香散中丁香酚和甘草酸的含量测定。

Abstract

OBJECTIVE To establish a HPLC quantitative method for the determination of eugenol and glycyrrhizic acid in Bawei Tanxiang San simultaneously. METHODS The separation was performed on an Agilent ZORBAX Eclipse SB-C18 column(4.6 mm×150 mm,5 μm with the mobile phase consisting of acetonitrile -5% acetonitrile(include 3% glacialacetic acid(27∶73. The flow rate was 1 mL·min -1 and the detection wavelength was at 280 nm(0~17.0 min, 250 nm(17.1~30 min. RESULTS The retention time of eugenol and glycyrrhizic acid was about 15 min and 21 min respectively. The resolution was more than 1.5. The regressive equation for eugenol was Y=0.106 4ρ-1.338(r=0.999 9,and the linear range was 77.76 -777.6 μg·mL-1,glycyrrhizic acid was Y=0.133 9ρ-0.222 0(r=0.999 9,and the linear range was 14.40~144.0 μg·mL-1 .The average recoveries of eugenol and glycyrrhizic acid were 99.0% and 97.3%, RSDs were 1.5% and 1.8%, respectively. CONCLUSION This method is simple,time-saving and accurate. It can be used for routine analysis of eugenol and glycyrrhizic acid in Bawei Tanxiang San.
