目的 采用新型缓释材料制备菊花提取物(CME)缓释片。方法 以总黄酮的释放度为指标,根据缓释片的有关参数采用正交试验、人工神经网络等方法优化处方。并以CME体内效应成分木犀草素、芹菜素的释放度为指标,用HPLC对处方优化后制备的缓释片进行释放度测定。结果 优化的CME缓释片的处方为:CME 150 mg,羟丙基甲基纤维素(HPMC)17 mg,1% 乙基纤维素(EC)乙醇溶液适量;以该处方制备的制剂,体外释药性能符合Mt/M∞=kt1/2模型,持续释药达12 h。结论 从体外释放分析,以该优化工艺制备的菊花缓释制剂,达到了缓释制剂的要求。
Abstract
OBJECTIVE To prepare the Chrysanthemum morifolium extract(CME) sustained-release tablets and study its releasing characterization in vitro. METHODS The orthogonal experiment and artificial neural network (ANN) were applied to optimize and predict the formulations of sustained release tablets. RESULTS The optimized prescription included 150 mg CME, 17 mg HPMC and 1% EC in one tablet. The tablets were releasing continually during 12 h and following Higuchi equation. CONCLUSION The in vitro release of CME sustained release tablets prepared as the optimized prescription is satisfied.
