摘要

目的 制备吲达帕胺有机酸复合物并对其透皮吸收进行研究。 方法 采用超声混合有机溶媒自然挥发方法制备吲达帕胺有机酸复合物,差式扫描量热法( DSC )测定吲达帕胺及有机酸复合物的熔点,红外吸收光谱法( FT-IR )对吲达帕胺有机酸复合物进行结构确认,用体外透皮吸收实验对吲达帕胺有机酸复合物的透皮吸收进行研究。 结果 DSC 和 FT-IR 图谱均证明吲达帕胺与有机酸之间形成了复合物,体外透皮吸收研究结果表明,吲达帕胺与有机酸形成复合物后能明显促进吲达帕胺的透皮吸收。 结论 由于吲达帕胺的透皮吸收量较低,可以采取与有机酸形成复合物的方法促进吲达帕胺的透皮吸收。

Abstract

OBJECTIVE To prepare indapamide and organic acids complexes and to evaluate the transdermal absorption of the complexes. METHODS Indapamide and organic acids complexes were prepared by ultrasound mixing method, and the solvent was natural evaporated. DSC method was used to determine the melting point of indapamide ant its complexes, and FT-IR method was used to identify the chemical structure. Using in vitro transdermal absorption experiments, the transdermal delivery properties of indapamide and its complexes were studied. RESULTS The spectrum of DSC and FT-IR indicated the formation of indapamide and organic acids complexes, and the result of in-vitro transdermal absorption experiments showed that accumulative amount of permeated indapamide was significantly (P<0.05) increased after the formation of indapamide and organic acids complexes.CONCLUSION Since indapamide has a low trandermal delivery amount, the preparation of indapamide and organic acids complexes may be a good method to promote the transdermal delivery of indapamide.
