OBJECTIVE To develop a sensitive RP-HPLC method for the determination of berberine chloride in beagle dogs,and to evaluate the duodenal targeting property of coated-bioadhesive tablets(CBT).METHODS The distribution of berberine chloride in lumina contents of GI tract was measured by HPLC after oral administration of CBT or equivalent dose of IRT in beagle dogs.RESULTS Area under the curve for CBT and IRT in duodenal content were 14.723 and 5.032 mg·h·g-1.The cmax of CBT and IRT were 4.630 and 2.345 mg·g-1.CONCLUSION Berberine chloride coated-bioadhesive tablets shows a good duodenal targeting property.
