摘要

目的探讨貂心肌线粒体DNA(mtDNA)的特征,建立中药利心丸中貂心的分子遗传学鉴定方法。方法从新鲜的貂心组织提取mtDNA,采用DNA限制性内切酶片段长度多态性(mtDNA-RFLP)分析技术和聚合酶链反应(PCR)扩增细胞色素b基因部分序列片段。结果新鲜的貂心组织可提取完整的mtDNA,并能扩增出310 bp大小的Cytb基因,与鸡心脏mtDNA限制性内切酶酶切图谱有差异。结论2种方法可以作为利心丸中貂心物种的区分提供方便有效的分子标记。

Abstract

OBJECTIVE To identifiy and characterize of mitochondrial DNA and cytochrome b gene in Martes zibellina L.heart and to establish the specific molecular technique for the purpose of protection,exploitation and utilization in LIXIN WAN.METHODS The mtDNA were extracted from fresh Martes zibellina L.heart and heart powder respectively,and the restriction fragment length polymorphism of mitochondrial DNA was used to construct their physical maps of mtDNA(mtDNA-RFLP) with 6 restriction endonuclease,the cytochrome b(Cytb) gene segment was amplified by PCR.RESULTS The complete mtDNA was separated from samples,and the Cytb gene with 310 bp segments was amplified by PCR.The restriction fragment length polymorphism of mitochondrial DNA gene with 6 restriction endonuclease were different between poultry and Martes zibellina L.heart.CONCLUSION The results reveal that both methods are usful for seeking out the genetic markers among the Martes zibellina L.heart and different species hearts effectively.
