摘要
目的研究不同比例黄连配伍对肉桂效应成分肉桂酸生物利用度的影响,探讨寒热并用复方交泰丸的药物配伍机制。方法将不同比例黄连配伍肉桂后灌胃小鼠,采用HPLC测定小鼠血清中肉桂酸的血药浓度,以单用肉桂为对照,计算不同比例配伍组成的复方中肉桂酸的相对生物利用度。结果肉桂酸在小鼠体内的药动学过程符合二室模型,不同比例黄连配伍肉桂后,交泰丸中肉桂酸的平均相对生物利用度分别为176.24%(肉桂-黄连=1∶2)、205.09%(肉桂-黄连=1∶4)。交泰丸与单药肉桂ρmax,AUC0~∞及tmax存在差异,其中交泰丸(肉桂-黄连=1∶4)ρmax,AUC0~∞最大。结论温里药肉桂与清热解毒药黄连配伍,能提高肉桂主要效应成分肉桂酸的生物利用度;且随黄连量的增加,肉桂酸的生物利用度相应提高。本实验从复方效应成分生物利用度的角度揭示了寒热并用复方配伍的科学内涵。
Abstract
OBJECTIVE To study the effect of Rhizoma coptidis on the pharmacokinetics and relative bioavailability of Cinnamomum cassia.METHODS The content of cinnamic acid in serum was determined directly by HPLC after an oral administration of Rhizoma coptidis in combination with Cinnamomum cassia.The serum concentration-time curves were plotted.The main pharmacokinetic parameters and relative bioavailability were obtained.RESULTS The serum concentration-time curve of cinnamic acid was discribed by a two-compartment model.The relative bioavailability of Rhizoma coptidis combined with Cinnamomum cassia was 176.24%(Cinnamomum cassia /Rhizoma coptidis =1/2),205.09%(Cinnamomum cassia /Rhizoma coptidis =1/4).CONCLUSION By the statistical analysis,Rhizoma coptidis combined with Cinnamomum cassia can increase the relative bioavailability of cinnamic acid.
