摘要
目的建立淫羊藿中绿原酸含量的毛细管区带电泳(CZE)测定方法,比较不同种的淫羊藿中绿原酸含量。方法采用熔融石英毛细管柱(50μm×95cm,有效长度86.8cm)作为分离通道;以20mmol·L-1硼砂(pH=9.12)为运行缓冲液;运行电压30kV;毛细管柱温20℃;检测波长:327nm;进样压力:5kPa,5s、外标法测定含量。结果淫羊藿中绿原酸成分峰得到完全分离,测定精密度得到明显改善;绿原酸的线性范围为3.28~42.64mg·L-1,r=0.9995;方法的加样回收率为100.36%,RSD为2.15%。结论本方法简便、准确,重复性好。
Abstract
OBJECTIVE To establish a capillary zone electrophoresis(CZE) method for the determination of chlorogenic acid in Epimedium brevicornum Maxim. and to compare the contents of chlorogenic acid in several different species of Epimedium brevicornum Maxim..METHODS A fused silica capillary(50 μm×95 cm,with effective length of 86.8 cm)was used.The running buffer was 20 mmol·L-1 borax(pH=9.12).The applied voltage was 30 kV and the capillary temperature was 20 ℃ with the pressure injection of 5 kPa×5 s.The detection wavelength was 327 nm.RESULTS The chlorogenic acid in Epimedium brevicornum Maxim.was separated completely, precision was improved obviously.The linear curve for chlorogenic acid was obtained in the range of 3.28~42.64 mg·L-1,r=0.999 5.The recovery was 100.36% with RSD of 2.15%. CONCLUSION The method is simple,accurate and good in reproducibility.
