摘要

目的建立高效液相色谱-紫外检测器测定球花石斛中滨蒿内酯和泽兰内酯含量的方法。方法采用ODS C18色谱柱(4.6 mmⅹ250 mm,5μm,MetaChem),以甲醇-四氢呋喃-1%醋酸水溶液(15∶5∶80)为流动相,流速为1 mL·min-1,检测波长为342 nm,柱温为40℃。结果球花石斛中滨蒿内酯的线性范围为0.004 64~0.464 00μg(r=0.999 9),平均回收率(n=6)为97.66%(RSD=0.27%);泽兰内酯的线性范围为0.004 24~1.060 00μg(r=0.999 9),平均回收率(n=6)为97.92%(RSD=0.89%)。结论本方法简便、准确、重复性好,可为评价不同产地的球花石斛质量提供一定依据。

Abstract

OBJECTIVE To develop an assay method for determining the concentrations of scoparone and ayapin in Dendrobium thyrsiflorum. METHODS High performance liquid chromatography-ultraviolet detection(HPLC-UV) was used.RESULTS The linear range of scoparone was 0.004 64~0.464 00 μg(r=0.999 9),and the average recovery(n=6) was 97.66%(RSD=0.27%);the linear range of ayapin was 0.004 24~1.060 00 μg(r=0.999 9),and the average recovery(n=6) was 97.92%(RSD=0.89%).CONCLUSION This method is simple,accurate and reproducible.It provide a basis for the quality evaluation for Dendrobium thyrsiflorum in different areas.
