目的:建立测定人血浆中多索茶碱浓度的反相高效液相色谱法。 方法:采用内标法,以μBondapak C18柱为固定相,甲醇-超纯水(38∶62)为流动相,咖啡因为内标,检测波长为275 nm,流速为1.0 ml·min-1,柱温:室温;血浆样品无需经过提取,直接沉淀蛋白离心后取样进行色谱分析。 结果:样品与内标的峰面积比同样品浓度间线性良好,线性范围在0.5~16.0 μg·ml-1,回归方程为:C=10.30R-0.18699,r=0.9992(n=6);平均回收率为99.9%(RSD=0.15%,n=5)。结论:该方法简单方便,快速,准确。
Abstract
OBJECTIVE To establish a method for determining the concentration of doxofylline in plasma. METHODS Plasma sample was used for the chromatography after protein precipitation and centrifuging on the basis of internal standard method, the μBondapak C18 column was used. Methanol-water (38∶62)(V/V) was used as mobile phase, and caffeine was used as internal standard, with the flow rate of 1.0 ml·min-1.The detection was carried out at 275 nm. The temperature of column was at room temperatur.RESULTS The separation and determination of the sample and internal standard could be accomplished preferably. The calibration curves were linear in the range of 0.5~16.0 μg·ml-1 for doxofylline .The average recoveries of doxofylline were 99.9%(RSD=0.15%,n=5). CONCLUSIONS This method provides a rapid and accurate way to measure the concentration of doxofylline in plasma.
