摘要

目的 寻找快速、准确、精密测定血浆同型半胱氨酸(HCY)浓度的方法;研究国内健康人体内HCY水平,及性别、年龄等因素对其影响。方法 用荧光偏振免疫法(FPIA)测定HCY血浆浓度。结果 FPIA法低、中、高质控日内RSD分别为0.61%,1.12%和1.52%,日间RSD分别为2.06%,2.54%和3.75%;回收率在(95.70±1.46)%~(99.90±1.11)%范围内。健康男、女性HCY水平分别为(8.76±2.81) μmol·L-1和(7.14±2.24) μmol·L-1(P=0.038)。年龄≤40岁组与年龄>40岁组,HCY水平分别为(7.27±1.98)和(9.25±3.24) μmol·L-1(P=0.016)。结论 FPIA法测定HCY水平准确可靠,方便易行;HCY水平与性别、年龄有关,男性明显高于女性,随着年龄的增长,HCY水平升高。

Abstract

OBJECTIVE To establish a method for the determination of homocysteine in plasma.To measure homocysteine levels in healthy adults in China.METHODS The fluorescence polarization immunoassay was used to assay the concentratin of HCY in plasma.RESULTS The relative standard deviation (RSD) of within-day varied from 0.61% to 1.52%,and inter-day from 2.06%~3.75% for the low,middle and high concentrations of check samples with the recoveries (95.70±1.46)%~(99.90±1.11)%,respectively.Homocysteine levels were (8.76±2.81) μmol·L-1 and (7.14±2.24) μmol·L-1in men and women respectively (P=0.038).Homocysteine levels were (7.27±1.98) and (9.25±3.24) μmol·L-1 in age ≤40 y and age >40 y respectively (P=0.015).CONCLUSION The levels in healthy Chinese individuals were similar to those reported abroad.FPIA method was rapid,accurate and sensitive for the determination of HCY in plasma.The gender difference in HCY concentrations was also significant.
