摘要

目的建立酒石酸唑吡坦血药浓度测定方法。方法采用HPLC-荧光检测法测定血药浓度,激发波长254 nm,发射波长390 nm。血样加0.25 mol·L-1KOH碱化后用乙醚萃取;色谱柱:HypersilODS2(200 mm×4.6 mm,5μm);流动相:乙腈-0.02mol·L-1KH2PO4(pH6.0)(40∶60);流速:1.0 mL·min-1。结果血药浓度线性范围5.0~250.0 ng·mL-1(r=0.9995,n=6),血浆最低检测浓度为2.5 ng·mL-1,高、中、低3种浓度平均回收率分别为:(103.60±2.44)%,(104.40±0.84)%,(106.64±9.93)%。结论该法为酒石酸唑吡坦药动学研究提供了一种检测方法。

Abstract

OBJECTIVE To establish a method for determining zolpidem tartrate in human plasma.METHODS A highperformance liquid chromatographic assay with fluorimetric detection was developed,the excitation and emission wavelengths were 254 nm and 390 nm,respectively.The plasma sample was extracted with diethyl ether after alkalization by addition of 0.25 mol·L-1KOH solution.The sample was separated on Hypersil ODS2 column (200 mm×4.6 mm,5 μm),with CH3CN-0.02 mol·L-1KH2PO4(pH6.0)(40∶60) as the mobile phase (flow rate at 1.0 mL·min-1).RESULTS The calibration curve was linear over the range of 5.0~250 ng·mL-1(r=0.9995,n=6).The limit of detection was 2.5 ng·mL-1.The mean recoveries of high,medium and low concentrations were (103.60±2.44)%,(104.40±0.84)%,(106.64±9.93)%,respectively.CONCLUSION This study provided a reliable quantitative method for the pharmacokinetic study of zolpidem.
