摘要
目的 测定胸腺肽溶液中α1的含量。方法 以胸腺肽α1纯品作为标准品,用高效毛细管电泳测定,未涂层石英毛细管柱(78 cm×30 μm);电泳缓冲溶液为0.05 mol·L-1磷酸盐缓冲液(pH 7.0);运行电压15 kV;负压进样(66.7 kPa);温度25℃;检测波长200 nm;进样时间1 s。用比例色谱的方法确定胸腺肽溶液中α1峰的纯度。结果 胸腺肽α1标准溶液的线性范围50~800 μg·ml-1,低、中、高3种浓度的平均回收率分别为87.2%,91.0%,97.4%。分离出的胸腺肽溶液中α1的组分峰为单一组分峰,3批胸腺肽溶液中α1的质量百分比分别为1.24%,0.812%,0.732%。结论 该方法可用于胸腺肽质控中α1含量的测定。
Abstract
OBJECTIVE To determine the content of thymosin alphal 1 in thymopeptide.METHODS High Performance Capillary electrophoresis (HPCE) analysis method was used at 25℃,using thymosin alphal 1 as the standard reference.The separation was performed on a 78 cm×30 μm uncoated sillica capillary column with the voltage 15 kV.The electrophoresis solution was 0.05 mol·L-1phosphate buffer (pH 7.0).The sample was detected with a UV-detector at 200 nm.RESULTS The linearity was obtained over the range of 50~80 μg·ml-1.The average recoveries of the check sample at the low,middle and high concentrations were 87%,91% and 97%,respectively.The mass content of thymosin alpha1 in 3 lots of thymopeptide solution were 1.24%,0.812% and 0.732% respectively.CONCLUSIONS The method could be used to determine the content of thymosin α-1 in thymopeptide for quality control.
