摘要

目的 从基因表达水平研究高山红景天苷对大鼠心脏内分泌功能的影响。方法 分别给各组大鼠按15 mg·kg-1·d-1ip红景天苷氯化钠溶液。饲养30 d后,断头处死。冷酚法提取心房总RNA,用(α32P)标记的大鼠心钠素前体原cDNA控针与各组大鼠心房总RNA做Northern blot杂交和Dot blot杂交,以检测心房ANP-mRNA的含量。结果 老年大鼠ANP-mRNA含量明显低于青年大鼠,注射红景天苷后,老年大鼠ANP-mRNA含量增加而青年大鼠减少。结论 老化过程中ANP基因表达能力明显衰退,红景天具有一定的抑制心脏内分泌功能老化的作用。

Abstract

OBJECTIVE To study the effects of gadoloside on atrial natriuretic peptide (ANP) gene expression.METHODS The rats were abdominally injected with sodium chloride solution of gadoloside 15 mg·kg-1·d-1for 30 days.Atria total RNA was extracted by the cold phenol method.The ANP-mRNA contents were determined using the Northem blot and Dot blot hybridization technique with α-32P-Labelled r-prepro-ANP-cDNA probe.RESULTS The ANP-mRNA contents of old rats were remarkably less than that of young rats.Salidroside increased the ANP-mRNA content of old rats,whereas it decreased the ANP-mRNA content of young rats.CONCLUSION The ANP gene expression declined during aging development and salidroside possessed anti-aging effects in the heart endocrine function.
