摘要
目的 建立反相高效液相色谱法测定人血浆中氢溴酸右美沙芬的浓度。方法 采用Nova-Pak C18色谱柱,以乙腈-水-36%乙酸(50∶49∶1),用三乙胺调pH至4.3,流速:1 ml·min-1,荧光检测,激发波长280 nm,发射波长310 nm。结果 氢溴酸右美沙芬约1.95 min达高峰浓度,cmax为(6.20±1.32)ng·ml-1。结论 本法简便、灵敏、专属性好,适用于氢溴酸右美沙芬血药浓度测定及药代动力学研究。
Abstract
OBJECTIVE To develop a RP-HPLC assay for the determination of dextromethorphan hydrobromide (DM) in healthy volunteers. METHODS The analytical colunm was packed with Nova-Pak C18.The mobile phase consisted of a mixture of acetonitrile-water-acetic acid (50∶49∶1).Triethylamine was used to adjust pH to 4.3.Flow rate was 1 ml·min-1.Fluor escence detector was used with Ex at 280 nm and Em at 310 nm. RESULTS:Tmaxws about 1.95 min.cmaxwas (6.20±1.32) ng·ml-1. CONCLUSION:The assay here was sensitive,simple and accurate,can be used to study pharmacokinetics of DM.
