摘要
目的:探讨表皮生长因子(EGF)在皮肤表面的吸收、透过及代谢?方法:采用氯胺-T法制备125I-EGF?采用放射性同位素直接测定法及三氯乙酸(TCA)沉淀法观察EGF不同时间累积透过量和透皮率及各时间段透过量和透皮率?结果:3 个剂量组12h累积透皮率均小于1%,EGF累积透皮率随涂抹剂量增大而减少,涂抹剂量与其透皮率呈非线性关系?结论:EGF透过皮肤时仅有极少量分解代谢,EGF作为局部外用药时透皮吸收量很少,可忽略作为外用药时全身吸收所产生的作用?本研究为今后的临床研究提供必要的参考资料?
Abstract
OBJECTIVE:To study the absorption,permeation and metabolism of epidermal growth factor (EGF) in the surface of the skin.METHODS:125I-EGF was labelled with Chloramine-T method and a Franz diffusion cell syntem was developed for the percutaneous absorption study.The accumulated permeation amount and ratio of 125I-EGF at the different time were determined by the direct radioisotope tracing assay and the trichloroacetic acid (TCA) precipitation method.RESULTS:The total accumulated permeability ratios during 12h were all less than 1% at the different dose of 125I-EGF.There was no linearity between the dose of 125I-EGF smeared on the surface of the skin and the accumulated permeability ratio. CONCLUSION:The study provided necessary imformation for further clinical trial.
