目的:建立高效毛细管电泳测定氯胺酮对映体血药浓度的方法。方法:血浆样品用三氯醋酸沉淀蛋白质,缓冲溶液为10mmol·L-1羧甲基-β-CD(CM-β-CD)加75mmol·L-1磷酸盐缓冲溶液,pH=5.00,紫外检测λ=210nm。结果:线性范围为2.5~25.0μg·ml-1,平均回收率为101.8%,日内和日间误差分别小于3%和5%。结论:本方法分析速度快,手性分离效率高,可用于开展临床研究。
Abstract
OBJECTIVE:To develep HPCE method for the determination of ketamine enatiomers in human plasma.METHODS:Plasma samples were handled by the addition of trichloroacetic acid to precipitate protein.The buffer was CM-β-CD-phosphate 7.5 mmol·L-1~75mmol·L-1with pH5.0.The detection wavelength was 210nm.RESULTS:The method was linear over the range of 2.5~25.0μg·ml-1.The average recovery was 101.8% and the validation of inter-day was less than 3% and 5%,respectively.CONCLUSION:This method is simple and rapid.The resolution of ketamine enantiomers is satisfied.It provides a suitable assay for clinical study.
