摘要

目的:探讨阿霉素(Adr)脂质体对小鼠肝癌-22细胞( H22)和肉瘤-180细胞( S180 )肿瘤的杭种瘤活性。方法:将Adr包封于二棕桐酸磷脂酰胆碱(DPPC)和大豆甾醇(SS)以及含有聚乙二醇(PEG)的两种脂质体(DPPC/SS和DPPC/SS/PEG脂质体)中,连同游离Adr,共3种样品,分别对接种了H22和S180的小鼠进行了尾静脉注射治疗实验。结果:Adr脂质休对H22腹水型肝癌和S180肉瘤的动物生命延长率和抑制效果优于游离的Adr,而且DPPC/ SS/PEG脂质体的杭癌效果更好。结论:将脂质体作为杭癌药物Adr的载体,对小鼠H22腹水癌和S180肉瘤的治疗有效。

Abstract

OBJECTIVE:To investigate the antitumor activity of adriamycin-liposomes (Adr-liposomes) against H22and S180 tumors in mice. METHODS: Adriamycin (Adr) was encapsulated in dipalmitoylphosphatidylcholine (DPPC) and soybean-derive}f sterol (SS) as well as PEG-DSPE liposomes (DPPC/SS-liposomes and DPPC/SS/PEG-liposomes).The two kinds of liposomes and free Adr were administered in the tail vein in H22 and S180 tumor-bearing mice. RESULTS: The antitumor efficacy of Adr encapsulated in DPPC/SS-liposomes and DPPC/SS/PEG-liposomes was active than that of the free drug and the DPPC/SS/PEG-liposomes were markedly active. CONCLUSION:These results indicate that liposomes delivery Adr may provide antitumor activity of Adr against H22 and S180 cancer.
