摘要

目的 建立HPLC检测厚朴温中汤水提液中甘草苷、橙皮苷、山姜素、6-姜酚、乔松素、和厚朴酚与厚朴酚的含量,为厚朴温中汤的质量控制提供参考依据。方法 采用色谱柱:北京慧德易SP ODS-AQ C18分析柱(4.6 mm×250 mm,5 μm);流动相:甲醇(A)-乙腈(B)-水(C);梯度洗脱;流速:1.0 mL·min-1;柱温:30 ℃;检测波长:290 nm;进样量:10 μL。运用HPLC法对厚朴温中汤的7个成分含量进行综合评价。结果 甘草苷、橙皮苷、山姜素、6-姜酚、乔松素、和厚朴酚与厚朴酚分别在2.92~146.00、2.28~114.00、4.26~213.00、4.12~206.00、2.64~132.00、1.82~91.00和4.00~200.00 μg·mL-1内与峰面积呈现良好线性关系(r=0.999 2~0.999 9),平均加样回收率(n=6)在98.4%~99.0%之间,RSD为0.68%~1.46%之间。结论 该方法学验证均符合要求,研究表明该方法稳定可靠、操作简便,可为厚朴温中汤质量控制及后期的标准汤剂工艺研究提供参考依据。

Abstract

OBJECTIVE To establish an HPLC method for detecting the contents of liquiritin, hesperidin, alpinetin, 6-gingerol, pinocembrin, honokiol and magnolol in Houpo Wenzhong Decoction water extract, and provide a reference for the quality control of Houpo Wenzhong decoction. METHODS SP ODS-AQ C18 (4.6 mm×250 mm, 5 μm); mobile phase: methanol (A)-acetonitrile (B)-water (C); gradient elution; flow rate: 1.0 mL·min-1; column temperature: 30 ℃; detection wavelength: 290 nm; injection volume: 10 μL. The content of 7 components of Houpo Wenzhong decoction was comprehensively evaluated by HPLC method. RESULTS The calibration curves of liquiritin, hesperidin, alpinetin, 6-gingerol, pinocembrin, honokiol and magnolol were in good linearity over 2.92-146.00、2.28-114.00、4.26-213.00、4.12-206.00、2.64-132.00、1.82-91.00、4.00-200.00 μg·mL-1 (r=0.999 2-0.999 9), respectively. The average recoveries (n=6) were in the range of 98.4%-99.0% and RSD were in the range of 0.68%-1.46%. CONCLUSION The methodological verification meets the requirements. The research shows that the method is stable, reliable, and easy to operate. It can provide a reference for the quality control of Houpo Wenzhong decoction and later standard decoction technology research.
