Exendin-4 induces ADSCs differentiation to osteoblasts into repair the bone defect

Abstract: Objective To determine the effect of Exendin-4 on bone regeneration in bone defect via the differentiation of ADSCs to osteoblasts. Methods We isolated the ADSCs and characterized them by flow cytometry. We divided mice into four groups: control, bone defect, bone defect+ADSCs and bone defect+ADSCs+Exendin-4. The Exendin-4 were given for 1 month to observe the bone formation. Results Exendin-4 induced ADSCs proliferation in dose- and time-dependent manner. Moreover, Exendin-4 promoted osteoblasts differentiation and inhibited adipocytes differentiation. Exendin-4 could also increase the rate of BV/TV, the trabecular number and thickness. Conclusion Exendin-4 promote osteogenesis in bone defect through osteoblasts differentiation of ADSCs.
