Abstract: Objective To study the effects of different high-dose X-ray on the expression of DNA damage repair related genes, the mortality and fertility in C.elegans. Methods Wild-type C.elegans were irradiated with 200 Gray (Gy) or 400 Gy of X-ray, then the expression of 14 DNA damage-repair related genes were quantified by RT-qPCR. STRING was adopted to analyze the protein-protein interaction of the differential genes.C.elegans survival was counted to assess the effect of X-ray on nematodes mortality and fertility. Results X-ray radiation caused abnormal expression of multiple DNA damage repair related genes in C.elegans as compared to the control group. Mortality of C.elegans increased(P<0.05)and fecundity decreased(P<0.05)after radiation. Conclusions X-ray radiation promotes apoptosis and inhibits fertility of C.elegans by changing the expression of multiple DNA damage and oxidation damage related genes. The age-1,cep-1,ced-4,rad-51 and daf-2 are sensitive to radiation, which are potential targets for radiotherapy.
