Abstract: Objective To identify whether human TRIM69 may inhibit the expression of cleaved capase7 and PARP in HeLa and HEK293T cells in response to UV irradiation or stimulation. Methods The gene of human TRIM69 was cloned into eukaryotic expression vectors, which was then transfected into HeLa or HEK293T cells. The cells were exposed to UV irradiation or stimulated with etoposide and then collected for western blot to detected the level of activation of caspase7 and the cleavage of PARP. Results The levels of expression of cleaved caspase7 and PARP showed a significant decline in the stable cells with ectopically expressed TRIM69 comparing with the control one (P<0.05). Conclusions Expressed TRIM69 inhibited the levels of expression of cleaved caspase7 and PARP in HeLa and HEK293T cells in response to both UV irradiation and etoposide stimulation.
