Antitumor effect of human amniotic epithelial cells supernatant on human hepatoma BEL-7402 cells in vitro

Abstract: Objective To investigate the migration, proliferation and cell apoptosis effects of human amniotic epithelial cells(hAECs) supernatant on human hepatoma BEL-7402 cells cultured in vitro. Methods BEL-7402 cells were randomly divided into five groups.The control group contained no hAECs supernatant, while the other four treatment groups were added different volume fraction of hAECs supernatant namely 6.25% ,12.5% ,25% and 50% for 24 hours.The scratch assay,MTT assay and flow cytometry were used to detect the migration, proliferation and apoptosis of BEL-7402 cells. The expression levels of apoptosis-related proteins were detected by Western blot method. Results hAECs supernatant could dose-dependently inhibit the migration and proliferation of BEL-7402 cells and induce apoptosis (P<0.05).The expressions of cleaved caspase-3 and cleaved caspase-8 of BEL-7402 cells in 25% and 50% hAECs supernatant groups were higher than those in control group(P<0.05). Conclusion hAECs supernatant has some antitumor effects on BEL -7402 cells cultured in vitro.
