Vitamin E succinate induces the apoptosis of carcinoma of esophagus cell line Eca-109

Abstract: Objective To study the effects of Vitamin E Succinate(VES) on proliferation, apoptosis and expressions of associated protein of human esophageal carcinoma cell line Eca-109.Methods Experimental groups were VES of different contrations and control group was culture medium in same volume. After Eca-109 cells were exposed for 24h,48h and 72h, MTT was used to measure the proliferation of cells.Morphological changes of Eca-109 cells were observed by transmission electron microscopy. Apoptotic rate and the expression of Survivin, Caspase-3 protein of Eca-109 among different concentration groups was examined by flow cytometry. Results The inhibitory rate increased gradually,with the increasing concentration of Vitamin E Succinate and prolonging of treatment time(P<0.01).Apoptosis morphous change appeared in different degree in cells. With the increasing concentration of VES,the apoptotic rate of every treatment group was significantly higher than that of control group(P<0.05). The protein expression of Survivin in Eca-109 reduced graduately(p<0.05),but Caspase-3 increased graduately(p<0.05). These two proteins had inverse correlation(r=-0.95, P <0.05). Conclusion: Vitamin E Succinate can inhibite the proliferation and induce apoptosis of Eca-109 cell line.The effect may be ralated to up-regulation the expression of Caspase-3 protein and down-regulation of the expression of Survivin.
