Influence of PABPC1 on translation activity in human monocytic leukemia cell line THP-1

Abstract: Objective To investigate the effect of cytoplasmic poly(A) binding protein-1(PABPC1) on translation of human monocytic leukemia cells line THP-1. Methods Specific short hairpin RNA(shRNA) was transfected into THP-1 cells. Western blot was used to verify the interference efficiency. The polysome profiling was used to examine the translation ability of THP-1 cells. Results The expression of PABPC1 protein was decreased in shPABPC1 transfected THP-1(P<0.05). Compared with control group, the translation ability of THP-1 cells in shPABPC1 group was reduced. Conclusions Knockdown of PABPC1 may inhibit the translation in THP-1 cells and PABPC1 is a potential target for the treatment of acute myeloid leukemia (AML).
