姜黄素抑制HeLa细胞端粒酶的活性

Curcumin inhibits telomerase activity in human cervical carcinoma Hela cells in vitro

摘要: 目的 研究姜黄素能否抑制HeLa细胞端粒酶活性。方法 用MTT法检测姜黄素对HeLa细胞生长的抑制作用,用RT-PCR和半定量TRAP-ELISA法检测HeLa细胞中人端粒酶催化亚基(human telomerase reverse transcriptase, hTERT)mRNA的表达和端粒酶活性。结果 HeLa细胞生长呈剂量依赖性抑制;其端粒酶活性和hTERT 的表达也明显下调。结论 姜黄素可能通过下调hTERT的表达而抑制HeLa细胞的端粒酶活性。

Abstract: Objective To investigate the inhibitory effect of curcumin on telomerase activity of HeLa cells. Methods The growth inhibition rate of HeLa cells treated with curcumin was measured with MTT method. The expression of hTERT mRNA was studied using RT-PCR. Telomerse activity was examined by TRAP-ELISA. Results The growth of HeLa cells treated with curcumin was inhibited in concentration dependent manner and the expression of hTERT mRNA and the level of telomerase activity were down-regulated significantly. Conclusion Curcumin can inhibit telomerase activity of HeLa cells.The underlying mechanism might be related to the down-regulation of hTERT transcription.
